Concurrent Enumeration of CD34+ and CD3+ Cells in Allogeneic Apheresis Collections Using a New Flow Cytometry Method

   

Donor lymphocyte infusions (DLI) are commonly used to prevent or treat relapse after allogeneic stem cell transplantation. DLIs may be collected through a separate apheresis procedure or derived from excess mobilized stem cell grafts. The latter approach is routine and requires quantification of both CD34+ stem/progenitor cells and CD3+ T lymphocytes. Currently, these measurements are performed in two separate assays by an independent laboratory.

We evaluated ACCELLIX™, a new flow cytometry method, that enables in-house simultaneous enumeration of CD34+ and CD3+ cells in a single assay with the potential to streamline graft characterization, reduce turnaround time, and decrease resource utilization.
