Summary

  • The Accellix Cell Phenotyping Platform automates sample preparation, voltage calibration, spectral unmixing, data acquisition, and data analysis.
  • T Cell and TBNK assay cell subsets were clearly and consistently identified by assay-specific autoclassification software across 30 unique leukapheresis samples, 3 lots of CD Chex Plus®, 15 Accellix instruments, and cell concentration levels ranging from 1 to 50 million cells per mL 
  • Coefficients of variance (CVs) for 5 replicates (5 instruments) across all samples ranged from 0.81% to 7.69%
  • Comparing manual and automated analysis results for each cell subset, the coefficients of determination (R2 values) are all at or above 0.9 and slopes range from 0.98 to 1.03, demonstrating excellent concordance and no systematic bias

 

The Accellix Platform has the potential to improve cell therapy QC efficiency by providing a simple and rapid workflow that minimizes process variability and subjective data analysis.
